Malattie rare – Paraplegia spastica ereditaria complessa causata da mutazioni in PCYT2

Abstract
CTP: la fosfoetanolamina citidililtransferasi (ET), codificata da PCYT2, è l’enzima di limitazione (determinazione) della velocità per la sintesi della fosfatidiletanolamina attraverso il percorso CDP-etanolamina.
La fosfatidiletanolamina è uno dei lipidi di membrana più abbondanti ed è particolarmente abbondante nel cervello.
Abbiamo identificato cinque individui con varianti PCYT2 bialleliche clinicamente caratterizzate da ritardo dello sviluppo globale con regressione, tetraparesi spastica o paraparesi, epilessia e atrofia cerebrale e cerebellare progressiva.
Utilizzando i fibroblasti dei pazienti, abbiamo dimostrato che queste varianti sono ipomorfe, provocano livelli di proteina ET alterati, ma residui e una concomitante ridotta attività enzimatica”, scrive il team.
I ricercatori sperano che un giorno queste firme possano essere utilizzate come biomarcatori per diagnosticare i pazienti con questa condizione.
Il team ha studiato gli effetti delle Paraplegie spastiche ereditarie (HSP) confrontando il pesce zebra con l’attività PCYT2 normale o ridotta e i campioni di cellule prelevati da pazienti affetti dalla malattia.
I ricercatori hanno scoperto che i tassi di sopravvivenza tra i pesci zebra con attività PCYT2 normale o ridotta erano significativamente più alti rispetto ai pesci con un knockout completo di PCYT2 .
